Well established gene disease association for multisystemic disease including kidney abnormality, muscle cramps, cerebral aneurysms, cardiac arrhythmia. Associated with small-vessel brain disease which may have varied severity and porencephaly with clinical manifestations of intellectual disability in ranging severity, motor delay, language impairment, seizures.
A few reports of homozygous variants in COL4A1 cause more severe phenotype (33912663; 33491999; 32042920)
