ID/DD is a feature of the condition

PLP1 is associated with PMD and hereditary spastic paraplegia SPG.
Different mechanism cause different disorders.
- Overexpression of either transcript leads to classical PMD (typically large or multi-gene duplications)
- Missense variants results in misfolding of both transcripts lead to severe PMD
- Null variants result in a milder present of PMD
