B-allelic variants in this gene cause CMT, Yunis-Varon syndrome and leukodystrophy, and mono-allelic variants are associated with ALS, and possibly with brain malformations.

Disease phenotype depends on the amount of residual FIG4 function.
Homozygous loss-of-function of FIG4 results in YunisVarón Syndrome. Partial loss-of-function of FIG4 can result in Charcot-Marie-Tooth disease (CMT) type 4J
